New type of ultraviolet light-sensitive mutation in yeast.
Four temperature-sensitive ultraviolet light-sensitive mutants of yeast have been isolated. All four mutations show complementation in diploid cells and the phenotypes associated with the rad (ts) mutations are the result of the activities of single mendelian genes.

Six uvr mutants of Saccharomyces cerevisiae with hypersensitivity to ultraviolet (UV) light were isolated after mutagen treatment with ethylmethanesulfonate. UV sensitivity ranges from moderate to extreme, and four of the mutants are also sensitive to nitrous acid. Ranking in terms of UV sensitivity does not parallel ranking in terms of nitrous acid sensitivity. Homozygous diploid mutant strain...

NDUCED mutations are thought to arise as a result of enzymatic processes uti‘lizing DNA damage as a substrate (BRIDGES 1969). Although the molecular mechanism is not known, evidence reviewed by WITKIN (1969) suggests that in E. coli mutations are produced during postreplication repair of lethal damage induced by ultraviolet light (UV) and controlled by r e d and ezrf (or lexf) genes. Strains ca...
متن کاملKinetics of mutation induction by ultraviolet light in excision-deficient yeast.
We have measured the frequency of UV-induced reversions (locus plus suppressor) for the ochre alleles ade2-1 and lys2-1 and forward mutations (ade2 adex double auxotrophs) in an excision-deficient strain of Saccharomyces cerevisiae (rad2-20). For very low UV doses, both mutational systems exhibit linear induction kinetics. However, as the dose increases, a strikingly different response is obser...
متن کاملUltraviolet Light Induced Mutation and Deoxyribonucleic Acid Replication in Bacteria.
HE terminal process through which the genetic change induced by ultraviolet Tlight (UV) is incorporated into the genetic apparatus is the initial postirradiation deoxyribonucleic acid (DNA) replication ( HAAS and DOUDNEY 1959; DOUDNEY and HAAS 1960). It has been suggested that two discrete radiation effects are prerequisite to incorporation of the “potential mutation” into DNA ( DOUDNEY, KADA a...
